Subscribe to our channel: https://www.youtube.com/@mkaeberlein Rapamycin has been called the most promising longevity drug ever discovered—and recently, some have claimed it's also one of the biggest disappointments. So which is it? In this deep dive, I separate evidence from hype and ask a simple question: Is rapamycin actually dead as a longevity intervention, or is the science stronger than ever? We explore what decades of research really tell us, from the landmark mouse lifespan studies that transformed aging biology to the latest clinical trials in humans and companion dogs. Along the way, I address many of the most common criticisms of rapamycin, discuss recent studies that have been widely misinterpreted, and explain why anecdotes and influencer opinions should never outweigh rigorous scientific evidence. Topics covered include: • Why rapamycin remains the most robust and reproducible longevity intervention in laboratory animals • What we know—and don't know—about rapamycin in healthy humans • Safety, dosing, and common misconceptions • Effects on immune function, brain aging, heart health, oral health, ovarian aging, muscle, and inflammation • What recent human clinical trials actually found • Why definitive longevity trials have been so difficult to perform • The future of rapamycin research, including ongoing clinical trials Rapamycin is not a miracle drug, and it is not proven to extend human lifespan. But after reviewing the totality of the evidence, I explain why I believe it remains the strongest scientific candidate we currently have for targeting the biology of aging. If you're interested in evidence-based longevity science—not hype—this lecture is for you.
