Can biological age actually be changed? A Yale-led analysis of 51 intervention studies says yes — and shows exactly which clocks respond.
Raghav Sehgal returns to walk through "DNAm aging biomarkers are responsive: Insights from 51 longevity interventional studies in humans" (Sehgal et al., Higgins-Chen lab).
The team built TranslAGE-Response, a harmonized database of 51 public and private longitudinal interventional studies, calculating a consistent set of 16 prominent epigenetic clocks for every study alongside 95 additional DNAm biomarkers that help explain what drives each clock's movement. Comparing intervention studies has been nearly impossible because every trial uses different clocks on different populations. This is the first time they've been put on common footing.
Three findings that matter most:
Clocks trained to predict mortality or pace of aging respond most strongly across all interventions — and they agree with each other. That is the opposite of the "different clocks say different things" problem that has held this field back.
Pharmacological and lifestyle interventions drive the strongest biomarker response, and study population and duration turn out to be key factors in whether an effect shows up at all.
If these biomarkers are genuinely responsive, they can act as surrogate endpoints — letting researchers monitor whether an intervention is working without running decade-spanning trials. The paper is explicitly built to help design better trials: which intervention, which subset of clocks to avoid multiple-testing problems, how long, what population, what sample size.
Why this matters beyond the lab: the clocks that respond most are the mortality-trained ones. Those are the same measures with demonstrated predictive power for mortality risk — which puts them squarely in the domain life insurers and reinsurers already underwrite against. If an intervention's effect can be read in months instead of decades, policyholders can benefit from positive feedback loops helping them to live longer, healthier lives.
