Senolytic drugs are a class of compounds designed to selectively induce the death of senescent cells. Senescent cells are aged or damaged cells that have stopped dividing but do not undergo apoptosis (programmed cell death). Instead, they accumulate in tissues and secrete pro-inflammatory cytokines, chemokines, and proteases—a phenomenon known as the senescence-associated secretory phenotype (SASP). The accumulation of these cells contributes to aging and age-related diseases.
Mechanism of Action:
Senolytic drugs target the pro-survival pathways that senescent cells rely on to resist apoptosis, including:
BCL-2 family proteins (e.g., targeting with Navitoclax)
PI3K/AKT pathway
p53/p21 pathway
Survivin
HIF-1α pathway
Examples of Senolytic Drugs:
Dasatinib (D): A tyrosine kinase inhibitor effective against senescent human preadipocytes and endothelial cells.
Quercetin (Q): A flavonoid with senolytic activity, particularly against senescent human endothelial cells and mesenchymal stem cells.
Navitoclax (ABT-263): A BCL-2 family inhibitor that induces apoptosis in senescent cells.
Fisetin: Another flavonoid with broad senolytic effects and lower toxicity compared to other senolytics.
FOXO4-DRI: A peptide that disrupts the interaction between p53 and FOXO4, promoting apoptosis of senescent cells.
HSP90 inhibitors: These interfere with cellular stress responses in senescent cells.
Potential Benefits:
Anti-Aging: Clearance of senescent cells may extend healthspan and potentially lifespan.
Therapeutic in Age-Related Diseases: Osteoarthritis, atherosclerosis, Alzheimer's disease, idiopathic pulmonary fibrosis, and more.
Cancer Therapy: As senescent cells in the tumor microenvironment can promote cancer progression.
Current Status:
Preclinical and Clinical Trials: Early human trials are showing promise in reducing senescent cell burden and improving clinical outcomes in age-related diseases.
Challenges: Balancing efficacy with safety, as broad senescence clearance could affect beneficial senescence, like wound healing or immune responses.
