NMN, the first molecular precursor to Nicotinamide adenine dinucleotide (NAD+), has recently attracted attention for its role in aging and lifespan extension. NAD+ directly and indirectly affects many cellular processes, including metabolic pathways, DNA repair, mitochondrial function, and immune cell activity. These mechanisms are critical for maintaining cellular homeostasis.
NAD+ concentrations begin to decline around 40 years old, just like klotho, which impairs tissue function, mitochondrial health, and a host of other functions critical to healthy ageing.
Many studies have looked at NAD+ metabolism and supplementation with its first two precursors, nicotinamide mononucleotide (NMN) and nicotinamide riboside (NR). Both supplements have shown to significantly increase intracellular NAD+ concentration. This is the opposite of what cyanide does. The opposite of instant death seems to me like it should be good.
As we age, the telomeres on the ends of our chromosomes get shorter. Shorter telomeres reduces cellular capacity for regeneration and increases risk of age-related impairment. Studies have shown telomerase activation can slow the progression of aging, and NMN and NR supplementation appear to activate telomerase, along with the anti-ageing suite of 7 Sirtuin genes.
