About this video

Creator description · original language

Subscribe to our channel: https://www.youtube.com/@optispan The R-Files is a series of episodes about rapamycin, a naturally occurring compound originally discovered in soil samples from Easter Island, also known as Rapa Nui (hence the drug's name). Rapamycin belongs to a class of drugs called macrolides and has potent immunosuppressive and anti-proliferative effects. The drug has garnered attention for its potential anti-aging properties and has attracted research interest for its ability to extend lifespan and delay age-related diseases in various model organisms, including yeast and mice. In this first episode of the R-Files, Matt, who has spent a significant chunk of his career studying rapamycin, will discuss how fortuitous conversations, an appetite for unexplored territory, and an opportunity to look where others weren't looking helped him get deep into the study of rapamycin and uncover some interesting insights into how rapamycin can affect lifespan. 0:00 Trailer 1:16 The beginnings of Matt's journey into studying rapamycin 4:07 Replicative aging in yeast 7:07 SIR4 gene as a regulator of yeast replicative lifespan 13:48 Unbiased genetic screens and how they can increase the validity of results 15:55 Why people may have thought Matt was crazy 18:25 Conducting an unbiased genetic screen of yeast replicative lifespan 19:00 TOR biology in yeast and how it relates to aging 22:53 The beauty of an unbiased genetic screen 24:38 TOR signalling and nutrient sensing 25:50 Why they didn't look at rapamycin at first 27:56 Testing the effect of rapamycin on chronological lifespan in yeast 29:12 A summary of the factors that combined to lead Matt to studying rapamycin 29:55 Expanding exploration in the longevity field today Producers: Tara Mei, Nicholas Arapis Video Editor: Jacob Keliikoa DISCLAIMER: The information provided on the Optispan YouTube channel is intended solely for general educational purposes and is not meant to be, nor should it be construed as, personalized medical advice. I spent the first few decades of my career doing scientific research into the biology of aging, trying to understand the finer details of how humans age in order to facilitate translational interventions that promote healthspan and improve quality of life. Now I want to take some of that knowledge out of the lab and into the hands of people who can really use it. On this channel I talk about all things aging and healthspan, from supplements and nutrition to the latest discoveries in longevity research. My goal is to lift the veil on the geroscience and longevity world and help you apply what we know to your own personal health trajectory.
