Rapamycin is one of the few compounds in aging research that has repeatedly delivered consistent results across an extraordinary range of species. In this video, longevity researcher Matt Kaeberlein explains why rapamycin remains the most reliable pharmacological intervention ever studied for extending both lifespan and healthspan. What makes rapamycin stand out is not hype, but biology. Its effects are tied to mTOR, a central cellular pathway that controls growth, metabolism, and aging. This pathway was first identified through unbiased genetic research aimed at answering a simple but powerful question: what actually controls lifespan at a fundamental level? Those early discoveries revealed something remarkable. The same biological mechanisms that regulate aging in yeast are often shared by worms, flies, mice, and mammals. Rapamycin taps directly into this conserved system. As a result, it has shown lifespan and healthspan benefits in every major model organism tested, including pet dogs. Importantly, the evidence does not rely on a single drug alone. Genetic inhibition of mTOR produces many of the same longevity benefits seen with rapamycin treatment. This dual line of evidence strengthens the case that mTOR plays a central role in biological aging itself, not just in individual diseases. The video also addresses a common misconception in longevity science: that simple organisms cannot teach us anything meaningful about human aging. In reality, decades of genetic research show that aging biology is deeply conserved across evolution. That conservation is what allows laboratory discoveries to translate into mammalian and clinical research. Together, these findings explain why rapamycin continues to anchor modern longevity science and why it remains at the center of discussions about healthspan, resilience, and aging biology.
