Most longevity drugs fail when tested long-term. Rapamycin keeps working. That’s why aging researchers continue to rank it as the most reliable intervention in longevity science. Rapamycin stands out in longevity research for one simple reason: it works consistently where most interventions fall short. While many compounds show early promise, very few demonstrate reliable effects across multiple studies and species. Rapamycin is one of the rare exceptions. In this video, leading aging researcher Matt Kaeberlein explains why rapamycin continues to earn serious attention from experts studying lifespan and healthspan. Research shows that rapamycin impacts the fundamental biology of aging rather than targeting a single disease. It influences pathways involved in cellular growth, repair, metabolism, inflammation, and resilience. One of the key reasons experts favour rapamycin is its broad reach. Evidence suggests it affects all major hallmarks of aging, including protein homeostasis, mitochondrial function, immune regulation, and cellular cleanup through autophagy. Instead of addressing symptoms downstream, rapamycin acts upstream at the level of biological aging itself. Rapamycin has been tested extensively in laboratory mammals, including mice and rats, where it repeatedly extends lifespan and improves multiple markers of healthspan. Importantly, its effects appear consistent across different genetic backgrounds, an area where other longevity strategies, such as caloric restriction, sometimes fail or even cause harm. This discussion also highlights why rapamycin is considered the most robust pharmacological intervention currently available in aging research. While it has not yet been tested as broadly in humans, its reproducibility in preclinical models makes it a cornerstone of modern longevity science. If you’re interested in how scientists think about slowing biological aging rather than treating individual diseases, this video offers a clear and evidence-based perspective on why rapamycin remains at the centre of the field.
