Why do human bodies age and die, and can science extend healthy life without creating new biological risks? In this COSMORA HQ AUDIO PODCAST, we explore the principal ideas of Why We Die: The New Science of Aging and the Quest for Immortality by Nobel Prize-winning molecular biologist Venki Ramakrishnan. Aging is not controlled by one biological clock, one gene, or one damaged molecule. It involves interconnected changes across DNA, proteins, mitochondria, metabolism, stem cells, immune function, cellular communication, and entire organs. Scientists can modify some of these processes in cells and laboratory animals. However, changing a biomarker or extending an animal’s lifespan does not prove that the same intervention will safely extend human healthspan, average lifespan, or maximum lifespan. In this independent BOOK OVERVIEW, we examine: • Evolutionary explanations for aging • The biological trade-off between reproduction, repair, and maintenance • DNA damage and declining genome stability • Telomeres and the limits of using telomere length as a measure of aging • Epigenetic changes and biological-age clocks • Ribosomes, protein production, and cellular quality control • Loss of proteostasis and damaged protein accumulation • Autophagy and cellular recycling • Mitochondrial function, energy production, and biological stress • Nutrient-sensing pathways, including mTOR and AMPK • Cellular senescence and its protective and harmful effects • Stem-cell exhaustion and declining tissue repair • Chronic inflammation and changes in immune function • The microbiome and its relationship with health and aging • Why different animal species have dramatically different lifespans • The difference between chronological age and biological age • Calorie restriction and the limits of current human evidence • Metformin, rapamycin, senolytics, and other experimental approaches • Partial cellular reprogramming and its scientific risks • Supplements, hormones, peptides, and unsupported longevity claims • Healthspan, lifespan, and maximum lifespan • The ethical and social consequences of substantially longer human lives THE BIOLOGICAL HALLMARKS OF AGING A widely used scientific framework identifies multiple interconnected hallmarks of aging: • Genomic instability • Telomere attrition • Epigenetic alterations • Loss of proteostasis • Disabled macroautophagy • Deregulated nutrient sensing • Mitochondrial dysfunction • Cellular senescence • Stem-cell exhaustion • Altered intercellular communication • Chronic inflammation • Dysbiosis These hallmarks are research categories rather than twelve completely independent causes. Changes in one process can influence several others. WHAT IS WELL SUPPORTED? Aging is biologically complex and is a major risk factor for numerous diseases. Cells possess extensive maintenance and repair systems, but these systems are not perfectly accurate or unlimited. Age-related changes affect DNA stability, protein quality, mitochondrial function, immune regulation, tissue repair, metabolism, and communication among cells. Regular physical activity, appropriate nutrition, restorative sleep, vaccination, tobacco avoidance, social connection, and responsible management of established health risks can support health and function across the lifespan.
