AT1-AA, miR-122-5p and podocyte senescence in preeclampsia: mouse and cell study
In this abstract, AT1-AA raised miR-122-5p, which repressed FOXO3/SOD2 and was linked to podocyte senescence and proteinuria in pregnant mice; inhibiting miR-122-5p reduced these effects. [1]

The key findings
The authors examined public clinical transcriptomic datasets and urine samples from preeclampsia patients, then combined human podocyte experiments with a pregnant mouse model infused with AT1-AA. [1]
In the mice, AT1-AA exposure caused severe hypertension, albuminuria and podocyte foot process effacement, with podocyte-specific senescence marked by nuclear p21 and a senescence-associated secretory phenotype. [1]
Systemic AntagomiR-122-5p attenuated the rise in maternal systolic blood pressure and reversed glomerular senescence, suppressed local inflammatory cytokines and restored podocyte ultrastructure in this model. [1]

Clinical data and pregnant mouse model
The authors first examined public clinical transcriptomic datasets and checked the results in urine samples from preeclampsia patients to find dysregulated microRNAs. They then combined laboratory experiments with human podocytes and a pregnant mouse model infused with AT1-AA, an antibody that activates the angiotensin II type 1 receptor. A reporter assay tested the regulatory cascade. The abstract does not report sample sizes, follow-up or how many patients and animals were studied.

Senescence findings and miR-122-5p inhibition
miR-122-5p was upregulated in preeclampsia patients. In the mice, AT1-AA exposure produced severe hypertension, albuminuria and effacement of podocyte foot processes, together with podocyte-specific senescence marked by nuclear p21 accumulation and a senescence-associated secretory phenotype. AT1-AA raised mitochondrial reactive oxygen species by upregulating miR-122-5p, which repressed the FOXO3/SOD2 antioxidant defense. Systemic treatment with the microRNA inhibitor AntagomiR-122-5p attenuated the rise in maternal systolic blood pressure and reversed glomerular senescence, suppressed local inflammatory cytokines and restored podocyte ultrastructure.

Abstract-only scope of the therapeutic claim
The abstract describes AntagomiR-122-5p as offering a therapeutic strategy that could ease maternal hypertension and protect the renal filtration barrier. These are findings from cells and an AT1-AA-infused pregnant mouse model, not demonstrated benefits in people. The abstract does not report doses, safety data, study duration or whether the clinical datasets and urine samples came from the same patients. Only the abstract was reviewed; the full text was not reviewed.

How to interpret this
Abstract only; the full text was not reviewed. Sample sizes, follow-up, doses and safety data are not reported. Cell and AT1-AA-infused pregnant mouse results do not establish benefits in people.
Based on the study abstract; the full paper, tables and supplementary data were not reviewed.
