Senescence-independent SASP in HNSCC: cell and mouse-free lab study

In CAL27 HNSCC cells, dsRNA stimulation triggered an NF-κB-dependent, autophagy-dependent secretory program without classic senescence markers; it activated macrophages via IL-6, and the TLR3-autophagy-NF-κB-IL-6 signature was validated as a prognostic biomarker in oral squamous cell carcinoma. dsRNA triggered SASP without senescence in CAL27 cells

The authors studied head and neck squamous cell carcinoma (HNSCC), which resists chemoradiotherapy and immune checkpoint inhibition. They stimulated CAL27 cells with the dsRNA mimic poly(I:C) and used chloroquine to inhibit autophagy. The stimulation produced an NF-κB-dependent secretory program they call senescence-independent SASP, without proliferative arrest or classic senescence marker expression. CXCL1 was identified as a candidate key mediator. These are cell-culture findings; the abstract does not report patient outcomes from this experiment.

IL-6 loop to macrophages; autophagy inhibition in cells

According to the abstract, this SASP subtype started an IL-6-mediated transcellular inflammatory cascade that activated SASP in tumor-associated macrophages and remodeled the immunosuppressive tumor microenvironment. Inhibiting autophagy suppressed this SASP, reduced IL-6 secretion and disrupted the tumor-macrophage inflammatory loop. The abstract describes in vitro functional assays and integrated transcriptomic analysis; it does not report human survival results for autophagy inhibition, so this is not evidence that patients benefit.

Prognostic signature validated in oral squamous carcinoma

Clinically, the abstract states that the TLR3-autophagy-NF-κB-IL-6 signature was validated as a reliable prognostic biomarker for oral squamous cell carcinoma, a form of HNSCC. Clinical specimen validation was part of the work. The authors propose this axis as a promising therapeutic strategy to improve chemoradiotherapy plus immunotherapy efficacy; that is a proposal, not a demonstrated treatment. The abstract does not give sample sizes, follow-up times or effect estimates, and the full text was not reviewed.
