Part 1: https://www.youtube.com/watch?v=06kXAJNZa0A&t=335s Part 2: https://www.youtube.com/watch?v=TVJNLXQeCg0&t=294s Part 3: https://www.youtube.com/watch?v=US2aaISId1U&t=1698s The R-Files is a series of episodes about rapamycin, a naturally occurring compound originally discovered in soil samples from Easter Island, also known as Rapa Nui (hence the drug's name). Rapamycin belongs to a class of drugs called macrolides and has potent immunosuppressive and anti-proliferative effects. The drug has garnered attention for its potential anti-aging properties and has attracted research interest for its ability to extend lifespan and delay age-related diseases in various model organisms, including yeast and mice. In this second episode of the R-Files, Matt, who has spent a significant chunk of his career studying rapamycin, discusses his own journey to using rapamycin off-label and how it all began with a persistent pain in his right shoulder that a doctor suggested might take a year to resolve. Matt conducted an n=1 experiment to test whether a short course of rapamycin might improve the age-related inflammation that was the likely cause of his shoulder issue. 0:00 Trailer 0:33 Recap of the R-Files episode 1 0:50 Introduction to Matt's personal experiments with taking rapamycin off-label 2:20 How "frozen shoulder" kicked off the beginnings of Matt's rapamycin journey 7:44 How Matt decided on his rapamycin dosage 9:41 The results of Matt's first rapamycin experiment 11:15 Matt's subsequent rapamycin cycles and his current cycle 12:47 Possible benefits of short rapamycin cycles Producers: Tara Mei, Nicholas Arapis Video Editor: Jacob Keliikoa DISCLAIMER: The information provided on the Optispan YouTube channel is intended solely for general educational purposes and is not meant to be, nor should it be construed as, personalized medical advice. Now I want to take some of that knowledge out of the lab and into the hands of people who can really use it. On this channel I talk about all things aging and healthspan, from supplements and nutrition to the latest discoveries in longevity research. My goal is to lift the veil on the geroscience and longevity world and help you apply what we know to your own personal health trajectory.
